Eukaryotic translation initiation factor 3F (EIF3F) is a newly discovered tumor suppressor. The aim of the current study was to establish a novel therapeutic strategy of targeting EIF3F for hepatocellular carcinoma (HCC). In vitro cell growth assays were carried out on four human hepatoma cell lines (HepG2, BEL7402, HCCLM3, and Bel7404). The effects of EIF3F small interfering RNA (siRNA) on the proliferation of the aforementioned cell lines were evaluated, with transfection of EIF3F siRNA being conducted in Huh-7 cells with either of the two lentivirus vectors, pHBLV-U6-CTX or pHBLV-U6-GFP. The sequence of siRNA was 5′-UACCUCAACCUCAUGGGGCAC-3′. HCCLM3 cells were injected in nude mice, with siRNA targeting EIF3F packaged by both pHBLV-U6-CTX and pHBLV-U6-GFP being administered to the xenograft tumor, and the effects of targeting EIF3F on tumor growth were evaluated. The proliferation of BEL7402 cells was inhibited by EIF3F siRNA, and targeting EIF3F exerted an inhibitory effect on the proliferation of HepG2 and Huh-7 cells. After transfection with the EIF3F siRNA vector, the tumors formed in the nude mice were smaller than those formed in the negative control group. In conclusion, targeting EIF3F by RNA interference is a feasible therapeutic strategy for liver cancer.
