Photoshop uses a layer-based editing system that enables raster image creation and altering with multiple overlays that support transparency. It’s extremely easy to get a bit of a head start on your images — even if you’ve only learned one or two features — by using the Pen Tool in the Toolbox.
Using the Pen Tool
Select a brush, pencil, rubber, or any other tool you’ve selected from the Toolbox. On the Tool Options bar, make sure that the Pen option is set to Active and that the Cap option is set to Side.
The Pen Tool works differently from other tools that you use to select objects on a canvas. Instead of dragging a selection window over an image, you click and drag with the Pen Tool to create the outline of an object, path, or path-like shape. The shape is stroked around the area of the image that you want to select.
Although the name Pen Tool may suggest that you can draw on a layer by clicking the Pen tool and drawing on that layer, it works differently from other drawing tools. The outline of your drawn shape is placed in a layer of the Photoshop document. This process is outlined in the following steps:
1. Choose Pen from the Toolbox and click the layer thumbnail on the left side of the screen.
2. Click and drag over the image on the right. A bounding box appears where you are clicking. Notice that there is a small black stroke or line that marks the bounding box of the drawing.
3. If you make a mistake and drag outside the bounding box, Photoshop erases everything that you drew and start you over. If you want to place text or a shape over the entire image, make a selection window over the entire image and put the text or shape on the layer.
The bounding box is transparent and can be altered as you paint the image. If you want to paint on the canvas without a bounding box, choose No Boundary on the Tool Options bar.

Finally, given the fact that positive selection of CD8+ T cells is carried out primarily by CD4+ T cells, which are overproduced in aplasia, it would be of great interest to test whether co-administration of IL-21 and IL-7 would induce positive selection of CD8+ T cells.
Conclusions {#Sec18}
===========
We have shown that IL-21 induces apoptosis of antigen-specific T cells in the presence of IL-7. This effect of IL-21 is probably mediated through its ability to drive cell cycle progression of the T cells and thereby enhance proliferation of antigen-specific T cells. This effect of IL-21, although initially described in an experimental model of cancer, holds great promise for the treatment of cancer patients.
