1. Open an image in Photoshop. In most cases, the person who took the picture made extra efforts to avoid using random or high-contrast colors. A black-and-white image is best for touch-ups. When using a photo of people, be mindful of their skin tone. You can lighten or darken a person’s skin with the Levels tool (described in Chapter 6).
2. Import the image’s background into a new layer. Most images will have some kind of background, such as sky, black paper, or white. A simple layer with the background color will be good for most touch-ups. It makes it easier to hide an error if you cover it with a layer mask and paint over it with a color.
3. Make a new layer. It’s always good to make a new layer for each step of a touch-up. It helps you track each step separately, as well as make sure you don’t accidentally apply the same changes to two different layers.
4. Using a brush or pen tool, paint over the glitch area. The area that needs touch-up will often appear quite dark. This is usually where either the pixels have been removed or the color has been altered.
5. Adjust the layer’s opacity. Select the layer, right-click on it, and choose Layer Mask from the menu that appears. The layer will appear as a solid white mask in the Layers palette. Make sure the layer is set to Overlay or Screen so you can see the effect of the mask. Adjust the Opacity setting to smooth out the glitch. It is usually best to start with 50% opacity and adjust it if needed. You can use any brush, pen, or mask tool for this step.
6. Save the image. Once the touch-up is complete, save the image.

To estimate the value of an equation, developed from the plasma creatine phosphokinase (CPK) response to a standard agent (phenylbutazone, PBZ) and validated on a sub-sample of blood donors, to predict the blood donors’ post-transfusion aminotransferases (ALT and AST) post-donation. Prospective, descriptive study in healthy dog blood donors. In a first step, the model was assessed in a sub-sample of donors (n = 24), using PBZ (0.2 mg/kg), a mild exercise protocol and on a blood sample taken at baseline and up to five hours post-donation. In a second step, the model was validated on the remaining blood donors (n = 112) using PBZ (0.3 mg/kg) and a strong exercise protocol. The blood donors had their aminotransferases measured post-donation (1-5 h) and were classified as responders and non-responders based on the increment of their AST and ALT values (>50 U/L). The model was developed using the data obtained from the sub-sample. The algorithm developed from the first step was then validated on the remaining blood donors. A 2 x 2 table was built using the PBZ results, with the blood donors’ responses (R) (0 = non-responders, 1 = responders) as the independent variable (X) and the predicted increments in the post-donation ALT (ΔALT) and AST (ΔAST) values as the dependant variable (Y). The performance of the algorithm was examined in terms of its sensitivity, specificity and predictive values. The algorithm was highly predictive for each aminotransferase, with an accuracy of 89% for AST and 93% for ALT. The model is highly predictive for both AST and ALT.
